mRNA (messenger RNA) screening as candidate for vaccines.

CleaScrip™ Custom mRNA (Low dsRNA)

Top-notch mRNAs characterized by  Yeasen's low dsRNA levels and exceptional integrity are indispensable for disease research utilizing mRNA tools and the advancement of RNA therapeutics.

https://www.yeabio.com/pages/cleascrip%E2%84%A2-mrna-synthesis-low-dsrna

Due to the ability of dsRNA to signal through Toll-like receptors TLR3 and RLR-mediated pathways, it is crucial to control the dsRNA content during clinical research. In primary cells and stem cells, the presence of dsRNA-binding receptors will inevitably trigger an immune response.

Yeasen offers exceptional control over the dsRNA content in mRNA.

In conventional mRNA synthesis services do not include a step for dsRNA removal. Adding this step increases costs, making the product more expensive. Typically, mRNA is categorized into RUO (Research Use Only) grade and preclinical grade, with dsRNA content being a key distinguishing factor. Yeasen Biotechnology, through collaboration with third parties and the use of the CleaScrip™ T7 RNA polymerase, can provide mRNA with ultra-low dsRNA content for drug or vaccine development scientists from the outset. This enables researchers to access near-clinical-grade quality mRNA at a lower cost from the very beginning.

SeekOne® Digital Droplet (SeekOne ® DD) Single Cell Whole Sequence Transcriptome Kit is a high-throughput single cell full sequence kit independently developed by Beijing Seek Biotechnology Co., Ltd. The kit breaks through the limitation that the traditional 3' or 5' transcriptome can only analyze the sequence at one end of the mRNA, and uses microfluidic digital droplets and Barcoded Beads conjugated with random primers to achieve random capture and detection of the transcriptome. The kit needs to be used with the company's self-developed SeekOne®® Digital Droplet System (SeekOne® DD) to complete the whole process from single-cell nucleic acid labeling to transcriptome library construction, and is equipped with single-cell data analysis software SeekSoul Tools to provide you with a one-stop single-cell transcriptome solution.

The SeekOne® DD Single Cell Whole Sequence Transcriptome Kit includes SeekOne® DD Chip S3 (Chip S3), Gasket, Carri-er Oil, SeekOne® DD Barcoded Beads (Barcoded Beads), amplification reagents, library construction reagents, and single-cell data analysis software (SeekSoul Tools).

A Single Cell Whole Transcriptome Sequencing Kit is a tool designed to analyze the full range of RNA transcripts from individual cells. Unlike bulk RNA sequencing, which pools RNA from many cells, single-cell transcriptomics allows researchers to explore the gene expression profiles of individual cells, revealing cellular heterogeneity, rare cell populations, and dynamic gene expression changes in tissues.

Here’s a breakdown of the kit’s components and features:

  1. Single-Cell Isolation and Capture: The kit typically uses microfluidic systems (droplet-based or plate-based) to isolate individual cells. Each cell is captured in a small droplet or well, keeping it separate for further processing.
  2. RNA Capture and Barcoding: Once isolated, RNA from each cell is captured and labeled with unique molecular identifiers (UMIs) or barcodes. This step ensures that the transcriptome from each cell is individually tagged, preventing cross-contamination between cells.
  3. cDNA Synthesis and Amplification: The captured RNA is reverse-transcribed into complementary DNA (cDNA), which is then amplified to increase the quantity of material available for sequencing. This allows even small amounts of RNA to be detected and analyzed.
  4. Library Construction: The kit includes reagents to prepare the amplified cDNA into sequencing libraries. These libraries are compatible with high-throughput sequencing platforms, such as Illumina or MGI, enabling large-scale parallel sequencing of thousands to millions of cells.
  5. Whole Transcriptome Coverage: Unlike targeted approaches that focus on specific genes, a whole transcriptome kit sequences all expressed genes, giving a comprehensive view of cellular gene expression. This is essential for discovering novel transcripts, non-coding RNAs, and alternative splicing events.
  6. Data Analysis: Many kits come with accompanying bioinformatics software or platforms, allowing researchers to analyze the high-dimensional data. These tools help identify differentially expressed genes, group cells into distinct types or states (like in cancer or immune studies), and explore complex biological pathways.

Applications:

  • Tumor Research: Identifying heterogeneous cancer cell populations, including rare tumor stem cells and metastatic cells.
  • Immunology: Profiling immune cells, understanding their roles in infection, autoimmunity, or therapeutic responses.
  • Developmental Biology: Mapping cell lineage and differentiation pathways during development.
  • Neuroscience: Investigating neuronal diversity and plasticity.
  • Infectious Diseases: Studying host-pathogen interactions at the single-cell level.
  • Drug Development: Exploring drug responses and resistance at a cellular resolution, allowing for more precise targeting in therapies.
